Isozymes as Genetic Markers in Citrus Growing in Cheju and Their Use for Identification of Nucellar and zygotic Seedlings


Abstract
Leaf protein extracts of Citrus growing in Cheju island were analyzed by polyacrylamide gel electrophoresis for isozymes of peroxidase (PX). The possibility of using the banding patterns of isozymes for the identification of nucellar and zygotic seedlings was examined. The results obtained are summarized as follows:
1. Seasonal changes in PX polymorphism were observed. PX isozymes, however, were considered to be used as genetic markers, since the trees of the same cultivar in the same season showed the identical PX zymograms.
2. Phenotypes of PX isozymes were identified in Citrus of 25 taxa and the related genera, but they were neither genetically explained nor correlated with taxonomic relations.
3. The lack of genetic explanation may limit the use of PX phenotypes in determining zygotic and nucellar seedlings.
